Lighting the iris of an eye

So I'm needing to film an ECU of an eye in a dark room and then have it transition to bright light as if the sun is coming up and is hitting the person's face. The camera will be extremely tight on the eye and I need to see the iris of the eye big and open when it's dark and get smaller as the light comes up.

 

I'm shooting on the 5D Mark ii and will most likely be renting a longer lens to really get in tight on the eye.

 

I am by no means a cinematographer but I've been learning a lot through this forum so any suggestions anyone has is totally welcome.

im assuming you are doing a shot like this one from "Requiem for a Dream"

 

Eye

 

Kino-flos with 216 in front of them work great for eye lights.... you can also take the bulbs out to make different designs. A 4x8 bounce board with a light shining into it also works really well
